If it's a genuine high quality card from a good manufacturer you'll probably be ok. I think inserting and removing the card is riskier than leaving it in the camera. Less chance to damage the card physically by accident, or misplacing it..
I would recommend getting another spare card for a vacation. I tend to like to have about twice the maximum capacity I think I will need. I also like to have at least one spare card just in case of failure. I've been on vacations where I've run out even after deleting many shots that aren't keepers. My largest cards are 4GB so I'm in a somewhat different situation than you with respect to capacity...

If the 16ghb card is from a name maker(sandisk or lexar), I would not worry about it. but if from one of the secondary super makers, maybe. remember anything make can break..
Third the idea, get a backup card of some size; I suggest 4gb or 2 of them..
I have 1 8gb card for emergency use. all the rest are 4gb or 2gb or 1gb. I have a grand totoal of 28gb of memory cards. since I shoot jpeg that is over 6900 images...
When I was confronted with buying memory I chose to buy several small 4G chips rather than larger chips because of the possibility of malfunction and I tought that I would be better organized if things were seperated that way...
Thanks to all of the above for their feedback. I unfortunately in the rush to enjoy a single large card got the first available in PNY, does anyone have any experience with that brand? I've used it for a little while now and it's worked fine for me, but I know not all cards are created equal. What is the probability that a card will breakdown if handled properly? I'm yet to have one go out, but previously I've been shooting on considerably smaller cards (a 2gb SanDisk actually)..

The truthfull answer is that it might and, then again, it might not. Experience tells me that losing one is more common and others will add that having them and the camera stolen is likely. But there's no way we can guarantee anything without a box of cards and a laptop to copy several times each hour... It's the NASA approach, btw..
Sorry I can't say more than that but I'll add that I use several small cards when I go on holiday and don't store the used ones with the camera as cameras are a target for thieves, especially in tourist areas..

PNY is one of the second tier makers. the 2 big ones are sandisk and lexar. I only get sandisk cards, IT SIMPLY IS NOT WORTH THE FEW DOLLARS TO TAKE A CHANCE. lexar is also very good...
